A Real Choice in STR Analysis

The PowerPlex® ESX and ESI Systems meet the ENFSI recommendation for DNA profile sharing across Europe. They are based on five-color technology and allow co-amplification and detection of D3S1358, D8S1179, D18S51, D21S11, FGA, TH01, vWA, D2S441, D10S1248, D22S1045, D1S1656, D12S391, D2S1338, D16S539, D19S433, SE33 and Amelogenin. These kits are available in multiple formats, including the option to detect SE33, to accommodate various requirements or preferences. Additionally, the kits have increased tolerance to common inhibitors and increased sensitivity to obtain full profiles from low-level DNA and are robust enough to genotype degraded DNA samples through the use of mini-STR loci.

The systems are compatible with the Applied Biosystems 3130, 3130xl and 3500 Genetic Analyzers and ABI PRISM® 3100, 3100-Avant and 310 Genetic Analyzers.

Consistent, Reliable Results

A DNA sample (0.5ng) heterozygous at all loci was amplified in a 25µl reaction with the PowerPlex® ESX 17 System for 30 cycles using a GeneAmp® PCR System 9700 thermal cycler. A loading cocktail containing CC5 Internal Lane Standard 500 and formamide was mixed with 1µl of amplified sample, and amplified fragments were analyzed using an Applied Biosystems 3130 Genetic Analyzer and a 3kV, 5-second injection. Data were analyzed using GeneMapper® ID software, version 3.2.1, and PowerPlex® ESX panels and bins files. Panel A. Fluorescein-labeled loci: Amelogenin, D3S1358, TH01, D21S11 and D18S51. Panel B. Exceptional Sensitivity

A DNA sample (60pg) heterozygous at all loci was amplified in a 25µl reaction with the PowerPlex® ESI 17 Pro System for 30 cycles using a GeneAmp® PCR System 9700 thermal cycler. A loading cocktail containing CC5 Internal Lane Standard 500 Pro and formamide was mixed with 1µl of amplified sample, and amplified fragments were detected using an Applied Biosystems 3130 Genetic Analyzer and a 3kV, 5-second injection.

A DNA sample (500pg) heterozygous at all loci was amplified in the presence of 300µM hematin in a 25µl reaction with the PowerPlex® ESX 17 System for 30 cycles using a GeneAmp® PCR System 9700 thermal cycler. A loading cocktail containing CC5 Internal Lane Standard 500 and formamide was mixed with 1µl of amplified sample, and amplified fragments were detected using an Applied Biosystems 3130 Genetic Analyzer and a 3kV, 5-second injection. Data were analyzed using GeneMapper® ID software, version 3.2.1, and PowerPlex® ESX panels and bins files.
