Advanced DNA Analysis for Forensic Applications

Enhanced Sensitivity and Speed

The VersaPlex® 31P System delivers rapid and reliable DNA profiling with the capability to complete amplification in under 80 minutes, ideal for urgent forensic investigations.

Advanced Discriminatory Power

The VersaPlex® 31P System features a 31-locus multiplex, integrating 20 CODIS core loci with 11 additional loci. The 11 additional loci include six novel autosomal markers and sex determination markers, ensuring superior analysis precision.

The 29 autosomal loci provide increased discriminatory power, while the two additional dye channels on the Spectrum and Spectrum Compact CE Systems result in 16 loci that have amplicons less than 250bp. This improves allele recovery from inhibited and degraded samples compared to similar 6-dye systems.

Loci configuration of the VersaPlex® 31P System.

Configuration of the VersaPlex® 31P System. The VersaPlex® 31P System allows co-amplification and detection of 31 loci including 20 CODIS core loci, Penta D, Penta E and D6S1043 and six novel autosomal loci (D6S477, D3S3045, D19S253, D10S1435, D15S659 and D8S1132). DYS391 and Amelogenin are included for sex determination, as well as two quality indicators (QIS) and (QIL) to assist with data interpretation. Sixteen loci in the VersaPlex® 31P System are less than 250bp, facilitating allele recovery from difficult or degraded samples.

The system is optimized to work with 0.5ng DNA input as shown in the electropherogram.

Electropherogram of amplifying extracted DNA with the VersaPlex® 31P System.

Amplifying extracted DNA with the VersaPlex® 31P System. 2800M Control DNA (0.5ng) was amplified using the VersaPlex® 31P System. Amplification products were mixed with CCO Internal Lane Standard 500 and separated on the Spectrum CE System. Electropherogram was generated using GeneMarker® HID Software for Spectrum CE Systems.

By including additional dye channels relative to the previous VersaPlex® multiplex, the VersaPlex® 31P System can give you improved and more informative analysis for even your most challenging DNA samples.

High Sensitivity and Specificity

The VersaPlex® 31P System is designed to generate complete DNA profiles with minimal sample input. Full profiles can be obtained with as little as 62.5pg of DNA, and an average of 88% of alleles can be called with just 31pg of DNA (n=8). This makes the system highly effective in cases involving degraded or limited samples, such as touch DNA or old forensic evidence.

Sensitivity of the VersaPlex® 31P System. Varying amounts of extracted DNA were amplified using the VersaPlex® 31P System. Amplification products were separated on the Spectrum CE System and analyzed using GeneMarker® HID Software for Spectrum CE Systems. The percent alleles called are an average of eight amplifications with each input DNA amount. The VersaPlex® 31P System demonstrates a sensitivity comparable to the PowerPlex® 35GY System, another Promega 8-color STR multiplex, showcasing the consistent performance of these systems.

Robust Amplification in Challenging Conditions

The robust amplification buffer in the VersaPlex® 31P System effectively overcomes common PCR inhibitors like hematin, tannic acid and humic acid, ensuring consistent, high-quality DNA profiles—even from challenging forensic samples. This reliability saves time and resources by reducing the need for repeat analyses.

Bar graph showing the robustness of the VersaPlex® 31P System.

Robustness of the VersaPlex® 31P System. Extracted DNA (0.5ng) in the presence of varying concentrations of PCR inhibitors was amplified using the VersaPlex® 31P System. Panel A shows results for no inhibitor, EDTA and hematin; Panel B shows results for humic acid and tannic acid. Amplification products were separated on the Spectrum CE System and analyzed using GeneMarker® HID Software for Spectrum CE Systems. The percent alleles called are an average of four amplifications with each inhibitor concentration. The VersaPlex® 31P System demonstrates resistance to inhibitors comparable to that of the PowerPlex® 35GY System when amplified under the conditions specified in the technical manual.
