NanoBRET® 590 Dyes Used in Early-Stage Drug Discovery Workflow
DNA-encoded libraries (DELs) provide unmatched chemical diversity and starting points for novel drug modalities. An article in Cell Chemical Biology presents a workflow that exploits the bifunctional attributes of DEL ligands as a platform to generate BRET probes for live-cell target-engagement studies. The combination of DEL and BRET technology accelerates hit-to-lead studies in a live-cell setting.

The NanoBRET® Target Engagement (TE) Assay measures compound binding at select target proteins within intact cells. This target engagement assay is based on the NanoBRET® System, an energy transfer technique designed to measure molecular proximity in living cells. The NanoBRET® Target Engagement Assay analyzes the apparent affinity of test compounds by competitive displacement of a NanoBRET® tracer reversibly bound to a NanoLuc® fusion protein in a cell.
We have a number of TE assays already developed for a large number of protein targets from a variety of protein classes. If there is no existing NanoBRET® TE Assay for your target of interest, use the NanoBRET® 590 Dyes to develop novel tracers to enable NanoBRET® TE assays for new targets.
How The NanoBRET® TE Assay Works
Illustration of the NanoBRET® TE Assay. Panel A. Compound engagement is measured in a competitive format using a cell-permeable fluorescent NanoBRET® tracer. Binding of the test compound results in a loss of NanoBRET® signal between the target protein and the tracer inside intact cells. Panel B. The affinity of the NanoBRET® tracer is determined for each target protein. For analysis of target engagement by a test compound, cells are treated with a fixed concentration of NanoBRET® tracer that is near the EC50 value of the NanoBRET® tracer dose-response curve. Panel C. To determine test compound affinity, cells are titrated with varying concentrations of the test compound in the presence of a fixed concentration (EC50–EC80) of the tracer.
Linker-Containing Dyes Accelerate Tracer Development
The NanoBRET® 590 Dyes can be used to develop novel cell-permeable tracers and are the same dyes used to create the NanoBRET® tracers available from Promega. The NanoBRET® 590 Dyes have consistently demonstrated robust cell permeability and optimal spectral overlap with NanoLuc® Luciferase for energy transfer. Three linker variants of the NanoBRET® 590 SE Dyes (Panels A–C) are available, allowing a panel of tracer candidates to be prepared from a common intermediate and providing the structural diversity needed to ensure the highest probability of success and, if required, to guide further optimization of tracer development. Also available is the NanoBRET® 590 Azide Dye (Panel D), which enables tracer synthesis via Azide-Alkyne Click chemistry and provides an amine orthogonal coupling chemistry.

NanoBRET® TE Assay Development Workflow
Workflow for developing a NanoBRET® TE Assay. The three stages of making a NanoBRET® TE Assay are: (1) develop a fluorescent tracer that will bind the target of interest; (2) construct a vector that expresses a fusion of the target protein with NanoLuc® Luciferase in cells; and (3) optimize the NanoBRET® TE Cellular Assay using NanoBRET® TE Assay reagents. NanoBRET® Dyes Limited Use Label License
BY USE OF THIS PRODUCT, RESEARCHER AGREES TO BE BOUND BY THE TERMS OF THIS LIMITED USE LABEL LICENSE. If researcher is not willing to accept the terms of this label license, and the product is unused, Promega will accept return of the unused product and provide researcher with a full refund.
Researcher may use this product for research use only, which shall include, without limitations, researcher’s (a) use of the product in its research and development of commercial products (provided that no such product requires the practice of Promega technology) (“Researcher Products”) or (b) use of any data generated using the product for the regulatory approval of such Researcher Products.
Researcher may not modify the product except to link the product to Promega or other non-Promega moieties (“Derivatives”).
For uses of this product and/or Derivatives for energy transfer (such as bioluminescence resonance energy transfer), researcher must:
(1a) use NanoLuc®-branded or NanoBiT®-branded luciferase, subject to any relevant label licenses, for all energy transfer determinations conducted with this product and/or Derivatives; and
(1b) use NanoBRET®-branded luminescent assay reagents (LARs; e.g., NanoBRET® Nano-Glo® Substrate, Intracellular TE Nano-Glo® Substrate/Inhibitor or Intracellular TE Nano-Glo® Vivazine®/Inhibitor) manufactured by Promega for all determinations of luminescent activity; or
(1c) contact Promega to obtain a license for use of the product or Derivatives with luciferases or LARs not manufactured by Promega.
Except as outlined in (2a) or (2b) below, researcher has no right to i) sell or transfer the product and/or Derivatives to another entity; (ii) sell data or information generated using the product and/or Derivatives to another entity; or iii) sell any product manufactured using the product and/or Derivatives to another entity.
(2a) If researcher is a contract research organization (“CRO”), such CRO may use the product solely on behalf of a single CRO customer to generate Derivatives and transfer or sell such Derivatives or data or information generated using the product and/or Derivatives to the single CRO customer, provided that CRO provides its single customer with a copy of this label license with the transfer of any Derivatives and the CRO customer agrees to comply with this label license for any use of the Derivatives; or
(2b) If researcher is not a CRO, researcher may transfer this product and/or Derivatives to a CRO in order to have CRO generate Derivatives solely on behalf of researcher or have CRO use Derivatives to provide researcher information, data or new Derivatives, and CRO may transfer the information, data or Derivatives to researcher, provided that researcher provides CRO with a copy of this label license and CRO agrees to comply with the terms of this label license.
With respect to any uses outside this label license, including any diagnostic, therapeutic, prophylactic, or commercial uses, please contact Promega for supply and licensing information. PROMEGA MAKES NO REPRESENTATIONS OR WARRANTIES OF ANY KIND, EITHER EXPRESSED OR IMPLIED, INCLUDING FOR MERCHANTABILITY OR FITNESS FOR A PARTICULAR PURPOSE WITH REGARDS TO THE PRODUCT. The terms of this label license shall be governed under the laws of the State of Wisconsin, USA.
