FuGENE® SI is an innovative, 100% synthetic, multicomponent transfection reagent designed for the delivery of siRNA, miRNA and other small RNA molecules. FuGENE® SI provides high-efficiency delivery of RNA into a wide range of eukaryotic cell lines while providing the gentle, low-toxicity transfection you expect from FuGENE® reagents. Gentle and Nontoxic siRNA Transfection
FuGENE® SI Transfection Reagent delivers gentle and nontoxic siRNA transfection. HEK293-GFP and NIH3T3-GFP cell lines (Cell Biolabs) were seeded in 96-well plates according to user guides and subsequently transfected with 0.2, 1.0 or 5pmol of GFP targeting siRNA or negative control (Dharmacon™, Horizon) along with 0.3µl of FuGENE® SI Transfection Reagent. Cells were then analyzed via flow cytometry 48 hours post-transfection to measure total cell viability. Graphs show the gentle and nontoxic transfection of siRNAs using FuGENE® SI.
Superior Knockdown Efficiency
FuGENE® SI outperforms Lipofectamine™ RNAiMAX in siRNA knockdown efficiency. HEK293-GFP and NIH3T3-GFP cell lines (Cell Biolabs) were seeded in 96-well plates according to user guides and subsequently transfected with 0.2, 1.0 or 5pmol of GFP targeting siRNA or negative control (Dharmacon™, Horizon) along with 0.3µl of FuGENE® SI Transfection Reagent or 0.3µl of Lipofectamine™ RNAiMAX (Life Technologies). Cells were then analyzed via flow cytometry 48 hours post-transfection to measure % knockdown of GFP and total cell viability. Graphs show the superior knockdown performance of FuGENE® SI versus Lipofectamine™ RNAiMAX in HEK293 and NIH3T3 cells.
FuGENE® SI outperforms Lipofectamine™ RNAiMAX in siRNA knockdown efficiency, as confirmed by Western blot. Human MDA-MB-468 cells were reverse-transfected in 6-well plates with 40pmol of the indicated targeting siRNAs or negative/positive control (Silencer® Select siRNA, Invitrogen) utilizing either 7.5µl of FuGENE® SI Transfection Reagent or 7.5µl of Lipofectamine™ RNAiMAX per well. The cells were collected 72 hours after transfection and analyzed via Western blot for expression of target genes STAT3 and CEBPB. The Western blot shown here demonstrates the robust, superior knockdown with FuGENE® SI as compared to the market leading competitor.
Efficient Transfection of Labeled Small RNAs
FuGENE® SI provides efficient transfection of labeled small RNAs. A549, COS-7, HEK293 and HeLa cells plated in 96-well microplates were transfected with 0.4µl of FuGENE® SI or a no-reagent negative control and various amounts of FITC-labeled negative control siRNA. Twenty-four hours after transfection, the cells were analyzed via flow cytometry for FITC-positive signal.
