Simple and Efficient Total Nucleic Acid Extraction from a Broad Range of Sample Types for Pathogen Testing​
The Maxwell® RSC Pathogen Total Nucleic Acid Kit, used with the Maxwell® RSC or Maxwell® RSC 48 Instruments, is designed for automated, benchtop extraction and purification of total nucleic acid from stool, sputum, bronchoalveolar lavage (BAL), plasma, urine, universal or viral transport media containing nasopharyngeal swabs, and Pap media containing cervical swabs. The extracted total nucleic acid is ready to use in a variety of amplification-based assays to detect human pathogens like bacteria, viruses and parasites. The Maxwell® RSC Instrument can process up to 16 samples, and the Maxwell® RSC 48 Instrument can process up to 48 samples, in parallel with the simple touch of a button. The preprogrammed Pathogen Total Nucleic Acid method on the Maxwell® RSC Instruments enables extraction of total nucleic acid from different sample types simultaneously in a single extraction run, maximizing laboratory time and efficiency.
The Maxwell® RSC Pathogen Total Nucleic Acid Kit protocol involves three main steps:
- Prepare sample lysate using simple sample-specific protocols.
- Add the prepared sample lysate to the Maxwell® RSC Cartridge (RSCB) and mix with the binding buffer already present in the first well.
- Load the prepared Maxwell® RSC Cartridge (RSCB) on the Maxwell® RSC or Maxwell® RSC 48 Instrument for automated extraction of high-quality total nucleic acid in less than 40 minutes.
The Maxwell® RSC Pathogen Total Nucleic Acid Kit contains all the required reagents along with convenient, prefilled cartridges for total nucleic acid extraction from all the supported sample types. A range of sample input amounts can be used for extraction, and the 100µl elution volume results in concentrated total nucleic acid. The Maxwell® RSC Pathogen Total Nucleic Acid Kit offers research laboratories an easy-to-use platform solution with built-in flexibility for consistent and reliable total nucleic acid extraction from different sample types for a variety of analytical workflows such as PCR, qPCR and RT-qPCR assays.

The Maxwell® RSC Pathogen Total Nucleic Acid Kit, used with the Maxwell® RSC Instruments, extracts total nucleic acid from a range of diverse sample types. The Maxwell® RSC Pathogen Total Nucleic Acid Kit works with a variety of sample types including stool (Panels A and B), sputum (Panel C), bronchoalveolar lavage (Panel C), plasma (Panel D), universal transport media (UTM; Panel E) or viral transport media (VTM; Panel F) containing nasopharyngeal swabs, Pap media containing cervical swabs (Panel G) and urine (Panel H). All sample types were spiked with known quantities of inactivated pathogens as indicated, and total nucleic acid was extracted from each specimen (n=8) using the Maxwell® RSC Pathogen Total Nucleic Acid Kit. Undiluted total nucleic acid extracted from each sample type and its tenfold dilution were used in a qPCR assay specific for the appropriate pathogen target. The extracted total nucleic acid shows no detectable inhibition in amplification-based assays for pathogen detection.
The Maxwell® RSC Pathogen Total Nucleic Acid Kit shows equivalent performance with both the Maxwell® RSC and Maxwell® RSC 48 Instruments. Total nucleic acid extracted from sputum specimens (n=8) using either a Maxwell® RSC or Maxwell® RSC 48 Instrument showed comparable results in RT-qPCR targeting SARS-CoV-2.
