Glutamine/Glutamate-Glo™ Assay Technical Manual

Literature # TM496

The Glutamine/Glutamate-Glo™ Assay is a bioluminescent assay for rapid, selective and sensitive detection of glutamine and glutamate in biological samples. In this two-step assay, glutamine is first converted to glutamate by glutaminase, followed by glutamate oxidation and NADH production coupled with a bioluminescent NADH detection system. In the presence of NADH, a pro-luciferin Reductase Substrate is converted by Reductase to luciferin, which is used in a luciferase reaction to generate light.

The luminescent signal is proportional to the amount of glutamate and increases until all glutamate is consumed, at which point a stable luminescent signal is achieved.
