IDseek® DNA Sequencing Kits for Forensic MPS

The IDseek® portfolio is a suite of Massive Parallel Sequencing (MPS) kits for forensic DNA analysis developed by NimaGen and distributed in the United States exclusively by Promega Corporation. All IDseek® panels are powered by Reverse Complement PCR (RC-PCR), a next-generation library preparation chemistry that performs indexing, adapter ligation and target amplification in a single closed-tube reaction. Because samples receive their Unique Dual Index in the very first PCR step, they can be pooled together before purification—eliminating the need for individual sample cleanup and significantly reducing both hands-on time and contamination risk.

The simplified 5-step workflow (prepare master mix → dispense and add DNA → PCR amplification → pool samples → purify and sequence) is compatible with Illumina MiSeq®, MiSeq FGx® and MiSeq i100 sequencing systems. With a suite of panels to choose from, the IDseek® portfolio delivers a scalable, validated solution for laboratories seeking to transition from capillary electrophoresis to MPS.

IDseek® MPS Consumables

IDseek® MPS consumables are purpose-designed to support the complete RC-PCR workflow.

• AmpliClean™ magnetic cleanup beads remove unincorporated primers and short fragments after PCR amplification and are functionally equivalent to AMPure® XP beads at a lower cost per reaction.
• Unique Dual Index (UDI) plates contain predispensed, dehydrated bar codes that are added directly to the amplification reaction in the first step of library preparation, enabling early indexing and eliminating the sample-swapping risk associated with post-PCR indexing.
• Single-probe-panel kits (OmniSTR™ Global, mYSTR™, OmniHap™, OmniSNP™, CombiSTR™ Plus) use standard U01–U04 plates.
• Dual-probe-panel kits (mtDNA HVR, mtDNA Full Genome) require the corresponding dual plates (U01D–U04D).
• Library Normalization Kits use a qPCR-based approach to equalize relative read depths across pooled samples before sequencing, ensuring consistent data quality across all samples in a run.
